p53 and Tumor Cell Glycolysis

Abstract
The classic activity of p53 in inhibiting tumor development includes inducing tumor cell cycle arrest,apoptosis,and senescence.Also,increasing evidence suggests that p53-mediated metabolic regulation plays a complex and critical role in tumor occurrence and development.The metabolic reprogramming caused by the elevated expression of key enzymes of aerobic glycolysis(Warburg effect)(hexokinase 2,phosphofructokinase-1,and pyruvate kinase M2)is one of the characteristics of tumor cells.It completes the occurrence,proliferation and invasion of tumor cells by satisfying their huge demand for antioxidants and synthetic metabolites.p53 can regulate these three key glycolytic enzymes in numerous and complex ways.This article reviews the role and mechanism of p53 in regulating the key enzymes of aerobic glycolysis,and briefly describes the roles of p63 and p73,members of p53 family,in glycolytic regulation.The aim is to connect p53 with glucose metabolism and tumor suppression,and provide ideas for potential mechanism research in this ongoing and rapidly developing field.关键词
