痛泻要方合四逆散颗粒对CUMS诱导的抑郁大鼠HPA轴及PI3K/Akt信号通路的影响
Effects of Tongxieyao Fang and Sini Powder on HPA Axis and PI3K/Akt Signal Pathway in Depressed Rats In-duced by CUMS

Abstract
Objective:To observe the effects of Tongxieyaofang and Sini Powder on HPA axis and PI3K/Akt signaling path-way in depression model rats induced by chronic unpredictable mild stimulation(CUMS).Methods:Thirty-six healthy Wistar rats were randomly divided into groups according to their body weight.They were set as blank control group,model group,Low,Medium and High-dose TCM treatment group,and western medicine treatment group,each with 6 rats.Except that the blank control did not give any intervention,the other groups used chronic unpredictable mild stimulus(CUMS)to establish depression models.After the completion of the model building,drug intervention was carried out for 4 weeks,the model group was given dis-tilled water,the traditional Chinese medicine treatment group was given stomach Tongxieyao Fang and Sini powder granules,and the western medicine treatment group was given fluoxetine.Sucrose Water Preference Test,after the end of the sample,the en-zyme-linked immunosorbent assay(ELISA)test adrenal corticosteroid releasing hormone(CRH),corticotropin(ACTH),serum corticosterone(CORT),norepinephrine.The contents of(NE),dopamine(DA)and 5-HT(5-HT);Western blot analysis(Western blot)was used to detect the expression of phosphorylated PI3K and Akt in the hippocampus of each group.Results:Compared with the blank control,CRH,ACTH,and CORT in the depression model group were significantly increased,NE,DA,and 5-HT were significantly decreased,and the expression of phosphorylated PI3K and Akt protein increased,and the difference was statistically significant.Significance(P<0.05);After the intervention of Jingtongxieyao Prescription and Sini Powder,com-pared with the model group,the content of CRH,ACTH and CORT in the traditional Chinese medicine group were reduced,and NE,DA and 5-HT all had Increased,phosphorylated PI3K and Akt protein expression decreased,the difference was statistically significant(P<0.05).Conclusion:Tongxieyaofang and Sinisan granules may play a positive role in prevention and treatment of depression model rats by affecting the HPA axis and PI3K/Akt signaling pathway.关键词
