健脾止动汤对抽动障碍模型大鼠纹状体快速放电中间神经元的影响
Effection of Jian-Pi-Zhi-Dong Decoction on Fast-spiking Interneurons in Striatum of TD Rats

Abstract
Objective:We detect the changes of FSIs contents in striatum of TD rats induced by IDPN by immunohistochem-istry,in order to provide neuroanatomical evidence of treating TD for Jian-Pi-Zhi-Dong Decoction.Methods:36 male SD rats were randomly divided into 4 groups according to body weight,namely model group,tiapride hydrochloride group,Jian-Pi-Zhi-Dong Decoction group and blank control group,with 9 rats in each group.In addition to the blank control group,the other three groups were injected intraperitoneally with IDPN(250mg/kg/D)to establish TD rat models.After successful modeling,the stereotyped exercise score and intragastric administration were carried out.The western medicine group was given tiapride hydro-chloride suspension(21mg/kg/D),and the traditional Chinese medicine group was given Jian-Pi-Zhi-Dong Decoction(16mg/kg/D),and the blank control group and the model group were given normal saline(10mg/kg/D).After continuous administration for 4 weeks,the stereotyped exercise score and body weight were measured.The number and average optical density of FSIS in striatum of rats in each group were analyzed by immunohistochemistry.Results:① body weight:the body weight of the model group was significantly lower than that of the blank group(P<0.01).At the end of 4 weeks after gavage,the body weight of rats in Jian-Pi-Zhi-Dong Decoction group was higher than that in model group(P<0.05).② Stereotyped motor score:on the 7th day of modeling,except for the blank group,if the score of each animal in each group was greater than 1 point,then the modeling was successful.At the end of 4 weeks after gavage,the score of tiapride hydrochloride group was lower than that of model group(P<0.05),and that of Jian-Pi-Zhi-Dong Decoction group was lower than that of model group(P<0.05),but there was no significant difference between tiapride hydrochloride group and Jian-Pi-Zhi-Dong Decoction group(P>0.05).③ Number of PV positive neurons:the number of PV positive neurons in the striatum of the model group was significantly lower than that of the blank group(P<0.01).The number of PV positive neurons in the striatum of Jian-Pi-Zhi-Dong Decoction group was more than that of the model group(P<0.05).There was no significant difference in the number of PV positive neu-rons between tiapride hydrochloride group and Jian-Pi-Zhi-Dong Decoction group(P>0.05).④ Average optical density:the average optical density of PV positive neurons in the model group was significantly lower than that in the blank group(P<0.01).The average optical density of PV positive neurons in the striatum of Jian-Pi-Zhi-Dong Decoction group was higher than that of the model group(P<0.05).There was no significant difference in the average optical density of PV positive neurons between tiapride hydrochloride group and Jian-Pi-Zhi-Dong Decoction group(P>0.05).Conclusion:Jian-Pi-Zhi-Dong decoction can reduce stereotyped motor in TD model rats,which is equivalent to the curative effect of tiapride hydrochloride,and can im-prove the adverse reaction of weight loss in TD rats.The decrease of PV positive neurons in striatum may be involved in the twitch behavior of TD rats.Jian-Pi-Zhi-Dong Decoction can improve the twitch behavior of rats by maintaining or repairing the damaged PV positive neurons and restoring their number and expression intensity.关键词
