Abstract

In order to explore an environmental method for mitigating"white pollution",a polyethylene-degrading bacteria strain PB1 was isolated by enriching and screening the inorganic salt medium with polyethylene as the sole carbon source from the soil covered with polyethylene film.The colony of PB1 was round with neat edge,moist and smoot surface.Additionally,the bacterium is gram strain positive and rod-shaped.The 16S rDNA gene sequence analysis showed that it had more than 99%identity with multiple Brevibacillus strains.Based on these characteristics,the strain PB1 was preliminarily identified as Brevibacillus sp.After co-cultivation of PB1 and polyethylene for 35 days,the weight loss rate of polyethylene membrane was 3.05%,and the bacterial fluid concentration(OD600)in the culture medium showed an increasing trend.Obvious pits and dents on the surface of degraded polyethylene diaphragm were observed under scanning electron microscopy and with filamentous cracks.According to the Fourier infrared transform spectrometer,the enzyme activity of laccase and manganese peroxidase were 0.125 iu·ml~(-1)and 0.126 iu·ml~(-1)at 35 d.It was proved that strain PB1 could degrade polyethene efficiently.Our study will enrich the strain resources of polyethylene-degrading bacteria and lay a foundation for the further study of polyethylene biodegradation.
