sPE患者血清miR-199a-5p、miR-142-3p表达变化及与产妇血管内皮损伤、围产儿结局的关系
Expression changes of serum miR-199a-5p and miR-142-3p in patients with sPE and their relationships with maternal vascular endothelial injury and perinatal outcome

Abstract
Objective To observe the expression changes of serum microRNA-199a-5p(miR-199a-5p)and microR-NA-142-3p(miR-142-3p)in patients with severe preeclampsia(sPE),and to analyze their relationships with maternal vascular endothelial injury and perinatal outcomes,so as to provide reference for the improvement of perinatal outcomes.Methods A total of 200 patients with sPE(observation group)and 200 healthy pregnant women(control group)were se-lected.The serum levels of miR-199a-5p and miR-142-3p were measured by RT-PCR.The serum levels of soluble endo-thelial growth factor(sEng),soluble vascular endothelial growth factor receptor-1(sFlt-1),endothelin-1(ET-1)and vas-cular endothelial growth factor(VEGF)were detected by enzyme-linked immunosorbent assay.Pearson correlation analy-sis was used to analyze the correlations between serum miR-199a-5p,miR-142-3p and vascular endothelial function-relat-ed indicators in the observation group.According to perinatal outcomes,the observation group was divided into the poor outcome group(29 cases)and good outcome group(171 cases).Univariate analysis and multivariate Logistic regression analysis were used to analyze the relationships between serum miR-199a-5p,miR-142-3p expression and perinatal out-comes.Results Compared with the control group,the expression levels of serum miR-199a-5p and VEGF levels de-creased,and the expression of serum miR-142-3p and the levels of sEng,sFlt-1 and ET-1 increased in the observation group(all P＜0.05).The expression of serum miR-199a-5p in the observation group was negatively correlated with the lev-els of sEng,sFlt-1 and ET-1,and was positively correlated with the level of VEGF(all P＜0.05).The expression of se-rum miR-142-3p was positively correlated with the levels of sEng,sFlt-1 and ET-1,and was negatively correlated with the level of VEGF(all P＜0.05).Significant differences were found in the age,the proportion of early-onset sPE,the propor-tion of concurrent edema,the proportion of concurrent eclampsia,the proportion of concurrent placental abruption,the proportion of concurrent postpartum hemorrhage,the expression of serum miR-142-3p,and the expression of serum miR-199a-5p between the poor outcome group and the good outcome group(all P＜0.05).Multivariate Logistic regression analy-sis showed that early-onset sPE,placental abruption and high expression of miR-142-3p were risk factors for poor perinatal outcome of sPE(all P＜0.05),and high expression of miR-199a-5p was a protective factor for poor perinatal outcome of sPE(P＜0.05).Conclusions The expression of serum miR-199a-5p in patients with sPE decreases and the expression of miR-142-3p increases,which are related to vascular endothelial injury.The high expression of miR-142-3p is a risk fac-tor for the poor perinatal outcome of sPE,and the high expression of miR-199a-5p is a protective factor for the poor perina-tal outcome of sPE.关键词
