自制鼻咽解毒胶囊含药血清对鼻咽癌细胞系5-8F存活、凋亡的影响及其机制
Effects and mechanism of self-made Biyan Jiedu capsule-medicated serum on survival and apoptosis of nasopharyngeal cancer cell line 5-8F

Abstract
Objective To observe the effects of self-made Biyan Jiedu capsule-medicated serum on the survival and apoptosis of nasopharyngeal carcinoma cell line 5-8F,and to explore its mechanism.Methods The 5-8F cells in the log-arithmic growth phase were divided into eight groups;cells in the 5%,10%,15%,20%,25%and 30%Biyan Jiedu cap-sule-medicated serum groups were added with 5%,10%,15%,20%,25%and 30%Biyan Jiedu capsules,cells in the 20%blank serum group were gavaged with 20%drug-free serum,cells in the control group were cultured normally;they were cultured for 24 and 48 h,and the cell viability of each group was detected by CCK-8.The 5-8F cells in the logarith-mic growth phase were divided into four groups;cells in the low-dose group were supplemented with 5%Biyan Jiedu cap-sule-medicated serum,the medium-dose group with 20%Biyan Jiedu capsule-medicated serum,the high-dose group with 30%Biyan Jiedu capsule-medicated serum,while cells in the control group were cultured normally for 24 h,and the apop-tosis rate was detected by flow cytometry.RT-qPCR was used to detect the relative expression levels of K-RAS and RAF mRNA in cells.Western blotting was used to detect the relative expression levels of K-RAS,RAF and p-ERK1/2 proteins in cells.Results Compared with the control group,the cell survival rates of the 20%,25%,and 30%Biyan Jiedu capsule-medicated serum groups decreased after 24 and 48-hour culture(all P＜0.05).Compared with the 20%blank serum group,the cell survival rates decreased in the 25%Biyan Jiedu capsule-medicated serum group after 48-hour culture and the 30%Biyan Jiedu capsule-medicated serum group after 24-and 48-hour culture(all P＜0.05).Compared with the same group at 24 h,the cell survival rate of the 30%Biyan Jiedu capsule-medicated serum group was reduced at 48 h(P＜0.05).Compared with the negative group,the apoptosis rates of the low-dose group,the medium-dose group and the high-dose group increased(all P＜0.05);compared with the low-dose group,the apoptosis rates of the medium-dose group and the high-dose group increased(both P＜0.05);compared with the medium-dose group,the apoptosis rate of the high-dose group increased(P＜0.05).Compared with the negative group,the relative mRNA expression levels of K-RAS and RAF in the low-dose group,medium-dose group and high-dose group decreased(all P＜0.05).Compared with the low-dose group,the relative mRNA expression levels of K-RAS and RAF in the medium-dose group and high-dose group decreased(all P＜0.05).Compared with the medium-dose group,the relative mRNA expression levels of K-RAS and RAF in the high-dose group decreased(both P＜0.05).Compared with the negative group,the relative expression lev-els of K-RAS protein,RAF protein and p-ERK1/2 protein in the high-dose group decreased,the relative expression levels of K-RAS and RAF proteins in the medium-dose group decreased,and the relative expression of RAF protein in the low-dose group decreased(all P＜0.05).Compared with the low-dose group,the relative expression levels of K-RAS,RAF and p-ERK1/2 in the high-dose group and the K-RAS and p-ERK1/2 proteins in the medium-dose group decreased(all P＜0.05);compared with the medium-dose group,the relative expression of K-RAS protein in the high-dose group decreased(P＜0.05).Conclusion The self-made Biyan Jiedu capsule-medicated serum can inhibit the survival of 5-8F cells and promote apoptosis in a dose-dependent manner,with the best effect in 30%Biyan Jiedu capsule-medicated serum,and the mechanism of action may be related to its inhibition of MAPK/ERK signaling pathway.关键词
