一株产表面活性剂菌株的筛选、培养条件优化及产物性质OACSTPCD
Screening,optimization of culture conditions and characteristics of its product of a surfactant producing strain
从长春市西郊污水厂底泥中筛选到一株能够产表面活性剂的细菌.该菌株菌落较小、呈乳白色,在显微镜下个体形态呈圆球状,为革兰氏阴性菌.16S rDNA鉴定结果表明:该菌株属于假单胞菌属,与Pseudomonas libanensis(KT767978)的相似度达100%,将其命名为Pseudomonas libanensis PbA.以葡萄糖为碳源,KNO3为氮源,在接种量质量分数为5%、27 ℃和pH 7条件下培养3 d,获得产物0.54 g/L.产物的荷电结果、薄层层析(TLC)分析及含糖量测定表明:该菌合成产物为糖脂类阴离子表面活性剂.产物的性质研究结果表明:当质量浓度为0.04 mg/mL时,其乳化指数为60.3%,排油圈直径为5.6 cm;表面张力从65.3 mN/m(质量浓度0.4 g/L时)减小至54.1 mN/m(质量浓度>106 mg/L时),临界胶束浓度(CMC)为106 mg/L.本研究为丰富生物表面活性剂生产菌,开发发酵生产流程及表面活性剂的应用提供了基础数据.
A bacterium capable of producing surfactant was screened from the sediment of sewage plant in the western suburb of Changchun.The colony of the strain is small and milky white.The individual shape is round under the microscope,and is gram-negative.The results of 16S rDNA sequence showed that the strain belonged to Pseudomonas sp.and was 100%similar to Pseudomonas libanensis(KT767978),was named Pseudomonas libanensis PbA.Product of 0.54 g/L could be synthesized by culturing the strain using glucose as carbon source and KNO3 as nitrogen source with the inoculation amount was 5%mass fraction at 27 ℃,pH 7 and cultured for 3 days.Analysis results showed that the synthetic product from the strain was glycolipid anionic surfactant by charged result,TLC analysis,and sugar content determination.The emulsification index and the diameter of oil drain ring of the surfactant with a concentration of 0.04 mg/mL was 60.3%and 5.6 cm,respectively.The surface tension of the surfactant decreased from 65.3 mN/mi(concentration 0.4 g/L)to 54.1 mN/m(concentration>106 mg/L)with the increase of concentration,and the critical micelle concentration(CMC)was 106 mg/L.The basic data are provided for enriching the surfactant praducing bacteria,opening the fermentation production process and the application of surfactants.
