七氟醚预处理和后处理对局灶性脑缺血再灌注损伤大鼠神经元坏死性凋亡的影响OA
Effects of Sevoflurane Pretreatment and Post-treatment on Necrotizing Apoptosis of Neurons in Rats with Focal Cerebral Ischemia-reperfusion Injury
目的 参照改良 Zea-longa 线栓法建立局灶性脑缺血再灌注损伤的大鼠模型,观察七氟醚处理对局灶性脑缺血再灌注损伤大鼠神经元的影响,并探讨其作用机制.方法 选取成年雄性SD大鼠进行实验,按随机分组原则分为4组:假手术(Sham)组、缺血再灌注(IR)组、七氟醚预处理(Spre)组、七氟醚后处理(Spo)组,每组各10只.造模结束后,采用Longa评分法评测神经行为学;酶联免疫吸附测定(ELISA)法检测大鼠脑组织中超氧化物歧化酶(SOD)活性及丙二醛(MDA)水平;采用Western blotting检测蛋白还原型烟酰胺腺嘌呤二核苷酸磷酸氧化酶 4(NOX4)、Caspase3的表达情况;流式细胞术检测细胞凋亡情况.结果 相对于Sham组,IR组的神经行为学评分、MDA水平、NOX4、Caspase3 的表达、神经元凋亡数明显升高,SOD活性降低,差异有统计学意义(P<0.05);与IR组相比,Spre组和Spo组的神经行为学评分、MDA水平、NOX4、Caspase3 的表达、神经元凋亡数明显降低,SOD活性升高,差异均有统计学意义(P<0.05).结论 在局灶性脑缺血再灌注损伤模型大鼠中,七氟醚预处理和后处理对缺血再灌注大鼠脑组织均有一定的保护作用,其保护作用可能通过抑制氧化应激损伤导致的神经元坏死性凋亡来实现的.
Objective To establish a rat model of focal cerebral ischemia-reperfusion injury by modified Zea-longa suture method,observe the effect of sevoflurane treatment on neurons of focal cerebral ischemia-reperfusion injury rats,and explore its mechanism.Methods Adult male Sprague-Dawley(SD)rats were randomly divided into 4 groups:Sham group,ischemia reperfusion(IR)group,sevoflurane pre-treatment(Spre)group and sevoflurane post-treatment(Spo)group,with 10 rats in each group.After the modeling,the Longa score method was used to evaluate neuroethology.Enzyme-linked immunosorbent assay(ELISA)was used to detect the activities of superoxide dismutase(SOD)and the levels of malondialdehyde(MDA)in rat brain tissue.Western blotting was used to detect the expression of NADPH oxidase 4(NOX4)and Caspase3 proteins.The cell apoptosis was detected by flow cytometry.Results Compared with Sham group,the neurobehavioral score,MDA levels,expressions of NOX4 and Caspase3 and neuronal apoptosis number in IR group were significantly increased,while the SOD activity was decreased,with statistically significant differences(P<0.05).Compared with IR group,the neurobehavioral scores,MDA levels,expressions of NOX4 and Caspase3 and neuronal apoptosis number in Spre group and Spo group were significantly reduced,while the SOD activity was decreased,with statistically significant differences(P<0.05).Conclusion In the modle of focal cerebral ischemia-reperfusion injury in rats,both pre-treatment and post-treatment with sevoflurane have certain protective effects on brain tissue of rats with ischemia-reperfusion injury.The protective effects may be achieved by inhibiting neuronal necrotic apoptosis caused by oxidative stress injury.
