摘要

Abstract

To explore the antibacterial activity of macrolides tilmicosin against intracellular Staphylococcus aureus(S.aureus),this study used S.aureus ATCC 29213 as the target bacterium and mouse monocyte macrophage cells(RAW264.7)as the host cells to construct an intracellular infection system of S.aureus.The growth curve of intracellular S.aureus was plotted,and the minimum inhibitory concentration(MIC),minimum bactericidal concentration(MBC),mutant prevention concentration(MPC),mutant selection window(MSW),post-antibiotic effect(PAE),and bactericidal curve of tilmicosin against intracellular S.aureus were determined.The results showed that intracellular S.aureus was in the lag phase,logarithmic phase,stationary phase,and decline phase at 0-4,4-18,18-24,and 24-48 hours,respectively.The MIC,MBC,MPC,and MSW were 4,8,12.8,and 4-12.8 μg/mL,respectively.The PAE and bactericidal curve results showed that tilmicosin exhibited a significant time-dependent killing characteristic against intracellular S.aureus.The results indicate that tilmicosin has strong antibacterial activity against intracellular S.aureus,providing a reference for developing rational dosing regimens for the treatment of bovine mastitis caused by intracellular S.aureus.
