Preliminary screening of biological phenotypes of single PilZ domain protein PA0012 and PA4324 mutants in Pseudomonas aeruginosa

Abstract
Objective This research aimed to study the effects of single PilZ domain cyclic diguanosine mono-phosphate receptor proteins PA0012 and PA4324 on biological phenotypes in Pseudomonas aeruginosa.Methods We first knocked out the genes encoding PA0012 and PA4324 by homologous recombination method,and ob-tained the mutant strains ΔPA0012 and ΔPA4324.Then,the wild-type strain PAO1 was used as the control to screen the phenotypes of the two mutants,including the ability to swarm,the ability to infect lettuce,the patho-genicity to nematodes and proteinase activity etc.One-way analysis of variance(one-way ANOVA)was used to analyze the differences among the groups.Subsequently,we performed RNA sequencing experiment to analyze the differential gene expression caused by the mutation of the two single PilZ domain proteins,and comprehen-sively analyzed the effects of these two proteins on biological phenotypes at the transcriptional level.Results In this study,we found that the mutation of these two single PilZ domain proteins could enhance the swarming motil-ity,weaken its ability to infect lettuce,and its toxicity to nematodes and protease activity did not change signifi-cantly in Pseudomonas aeruginosa.RNA sequencing results showed that mutations in both proteins could lead to the differential expression of a large number of genes in Pseudomonas aeruginosa,and there were some common differential genes.Conclusion Both PA0012 and PA4324 mutations of Pseudomonas aeruginosa can enhance their movement and reduce their infection to lettuce,which lays a foundation for the follow-up mechanism study.关键词
