Research on Data Processing Methods for Portable Real time Fluorescence Quantitative PCR Instrument

Abstract
Real time fluorescence quantitative PCR is increasingly being used to quantify the expression level of nu-cleic acids.With the development and progress of the economy and society,individuals'self-protection awareness and personal health have become more important,which has led to higher demands for health.The demand for on-site nucleic acid analysis technology is becoming increasingly significant,and different instruments have different robust and suitable measurement and analysis methods.This study focuses on the data processing and analysis of a portable real-time fluorescence quantitative PCR instrument based on turntable fluorescence detection.By compa-ring and optimizing different peak fitting algorithms and amplification curve fitting algorithms,the detection per-formance of the optical system has been improved.The results show that the peak fitting algorithm for the system uses sine fitting to process the original data has a good effect.The amplification curve fitting algorithm uses 4-pa-rameter logistic fitting.The linear Goodness of fit of the amplification results of standard concentration gradient samples is R2>0.990,and the CV value of the coefficient of variation is less than 5%.This improves the detection performance of the optical system,and provides some reference for the data processing methods of this type of de-tection model instrument.关键词
