Abstract

Aims:The physical and chemical properties and in vitro functional activities of Wuniuzao dark tea polysaccharides(WDTP)and its degradation products were investigated.Methods:WDTP was prepared by the hot water extraction method,and its degradation products were obtained by the free radical degradation method.The physical and chemical properties of WDTP and its degradation products were analyzed by ultraviolet spectroscopy,high performance liquid chromatography and high performance gel chromatography.The antioxidant activity of WDTP and its degradation products in vitro was compared by the DPPH free radical scavenging assay,the ABTS free radical scavenging assay,the hydroxyl free radical scavenging assay and the potassium ferricyanide method.The lipid-lowering activity of the WDTP and its degradation products in vitro was compared by the pancreatic lipase inhibition assay and the lipid accumulation assay in HepG 2 cells.Results:After free radical degradation,the content of WDTP increased from 63.74%to 65.28%.The content of uronic acid decreased from 17.15%to 14.67%.The composition of monosaccharides was the same.The molar ratio of monosaccharides was slightly different;and the weight average molecular weight of WDTP decreased from 67 303 u to 55 423 u.Compared with WDTP before degradation,the scavenging activities of DPPH radical,ABTS radical,hydroxyl radical and the total reducing power of WDTP degradation products were significantly enhanced.WDTP degradation products inhibited pancreatic lipase activity and lipid accumulation in HepG2 cells by increasing the mRNA expression levels of PPAR-a,CPT-1A and CYP7A1 genes.Conclusions:WDTP has changed its physical and chemical properties after free radical degradation;and its antioxidant and lipid-lowering activities are enhanced in vitro.
