Abstract

To investigate the relationship between iNOS gene polymorphism and Brucella susceptibility in Kazakh sheep,(primers were designed for exons 15,16,17 and their adjacent introns according to the iNOS gene sequences in GenBank and the target gene fragments were amplified by PCR.SSCP gel electrophoresis was used to detect different genotypes and DNA sequencing was used to detect polymorphism for analysis of relationship between SNPs and Brucella Kazakh susceptibility.Total 67 sheep were found positive,accounting for 29.00%.Two polymorphic loci F15-C29002T and F15-G29076a were found in exon 15 fragment,f16-A30045G locus in exon 16 fragment and f17-T31213G locus on exon 17 fragment of iNOS gene of Kazakh sheep.Chi-square test showed that polymorphic loci f15-C29002T and F15-G29076A had no significant correlation with Brucella susceptibility(P>0.05)while F16-A30045g and F17-T31213g had very significant correlation with Brucella susceptibility(P<0.01).In conclusion,polymorphic loci F15-C29002T and F15-G29076A of iNOS gene of Kazakh sheep had no correlation with Brucella susceptibility while F16-A30045G and F17-T31213G had correlation with Brucella susceptibility.
