一株H1N1亚型猪流感病毒的分离鉴定及小鼠致病性的研究
Isolation,Identification and Pathogenicity in Mice of A H1N1 Subtype Swine Influenza Virus

Abstract
To elucidate the biological characteristics and pathogenicity of the A/Swine/Jiangsu/1070/2019(H1N1)virus isolated from a slaughterhouse in Jiangsu,the virus was purified by the chicken embryo limiting dilution method and then sequenced for genetic evolution analysis.The HA cleavage site of this isolate was 339PSIQSR↓G347,which conformed to the characteristics of low pathogenic SIV.The homology analysis found that the HA gene was 98.94%identical to A/Swine/Jiangsu/J006/2018(H1N1),and the NA gene was 98.79%homologous to A/Swine/Shandong/LY142/2017(H1N1).The internal genes were highly similar to the strains isolated in 2018.These results implied that A/Swine/Jiangsu/1070/2019(H1N1)was a recombinant virus in swine.A/Swine/Jiangsu/1070/2019(H1N1)replicated well on chicken embryos(108.5 EID50/0.1 mL)and MDCK cells(105.22 TCID50/0.1mL).The animal infection assay showed that the virus caused weight loss of the inoculated mice and the mortality was 33.3%on the day 6-7 post-infection.Moreover,the virus replicated in the lungs and hearts of the inoculated mice with titers reaching 2.91±0.19 lg EID50/mL and 2.37±0.29 lg EID50/mL,respectively.After infection,the pro-inflammatory cytokines IL-6,IL-12 and TNF-α were significantly up-regulated(P<0.01)and caused obvious pathological lesions in lungs.These results indicated that the isolated virus was a new reassortant and pathogenic to mice,which provided a certain reference for the early warning,prevention and control of H1 subtype swine influenza.关键词
