Abstract
In order to evaluate the combined anesthetic effect of bromadiolone and chloral hydrate in rats and mice,and explore an anesthesia method that is sufficient and has a suitable reversal agent for rats and mice.In this study,bromadiolone and chloral hydrate were co-administered by intraperitoneal injection,and the anesthetic responses in rats and mice were observed.Changes in heart rate,blood pressure,and body temperature in rats during anesthesia were monitored.After anesthesia,rats were injected with the reversal agent atipamezole into the muscle,and its awakening effect and impact on cardiovascular dynamics were observed.Mice were injected with atipamezole into the muscle after anesthesia,and the awakening effect as well as the impact on body temperature and bladder filling were observed.The results showed that the induction time of anesthesia in rats with 9 mg/(kg·bw)bromadiolone and 100 mg/(kg·bw)hydrated chloral hydrate was(4.0±1.4)min,the maintenance time was(27.8±13.3)min,and body temperature and blood pressure remained stable during anesthesia,but the heart rate decreased at 30 and 45 min after administration.A dose of 0.1 mg/(kg·bw)atipamezole could rapidly awaken rats and restore heart rate.In mice,the induction time of anesthesia with 12 mg/(kg·bw)bromadiolone and 250 mg/(kg·bw)chloral hydrate was(6.4±1.9)min,and the maintenance time was(9.4±2.7)min.A dose of 1.6 mg/(kg·bw)atipamezole could rapidly awaken anesthetized mice and avoid hypothermia and urinary retention.The results indicate that the combination of bromadiolone and chloral hydrate for anesthesia in rats and mice is a simple and effective method with fast onset,stable effect,and a suitable reversal agent.关键词
