Abstract

Objective:To establish in vitro rapid propagation system of Curcuma wenyujin introduced from Hainan Province,and to lay a foundation for factory seedling cultivation.Methods:The rhizome of Curcuma wenyujin in Hainan area was pretreated.Using bud as explants,the primary culture,secondary proliferation,rooting and transplanting of tissue culture seedlings were carried out,and the me-dium of each culture stage was optimized.Results:After soaking the rhizome with 10 mg/L GA3 for 30 min,the germination rate and germination rate of the Curcuma wenyujin rhizome which had been stored for half a year were increased.The contamination rate of rhizo-me germination in the initial culture was less than 5%,and the bud induction rate was 100%.The subculture medium was MS with 4 mg/L ZT and 0.4 mg/L LAA,and the proliferation coefficient was 2.4.The survival rate of tissue culture root seedlings was more than 93%with light matrix mesh bag container.Conclusion:The rapid propagation system was established in vitro by taking the rhizome of the introduced species of Curcuma wenyujin from Hainan as the explants,which can be applied to the factory breeding of tissue culture seedlings of Curcuma wenyujin,ensure the supply of high-quality seedlings,and promote the sustainable development of Curcuma wenyu-jin industry in Hainan.
