低温弱光下普通丝瓜的生理响应及转录组测序分析
Physiological Response and Transcriptome Sequencing Analysis of Luffa cylindrica under Low Temperature and Weak Light

Abstract
The seedlings of Luffa cylindrica cultivar'Nongfu 806'were treated with low temperature and weak light stress at 10℃and 5℃,respectively.Physiological parameters,photosynthetic parameters and chlorophyll fluorescence parameters were determined.Correlation analysis,principal component analysis and transcriptome sequencing were used to study the physiological response mechanism of Luffa cylindrica seedlings to low temperature and low light stress.The results showed that During low light treatment at 5℃(0~24 h),the activities of peroxidase(POD),catalase(CAT),superoxide dismutase(SOD)and the contents of malondialdehyde(MDA)and free proline(Pro)of Luffa cylindrica seedlings increased first and then decreased,while the content of soluble sugar increased in general.During low light treatment at 10℃(0~10 d),POD activity,MDA and soluble sugar content of Luffa cylindrica seedlings showed an increasing trend in general,and the other three indexes showed an increasing trend first and then decreasing trend in general.The content of soluble sugar in luffa seedlings was positively correlated with POD and SOD activities and negatively correlated with CAT activities.Pro content was significantly negatively correlated with MDA content.SOD activity was negatively correlated with CAT activity and positively correlated with POD activity.Principal component analysis was performed on 6 physiological indexes,and 3 principal components were extracted at 5℃and 2 principal components were extracted at 10℃.Transcriptome sequencing obtained 713 million Clean data.Among the differentially expressed genes of luffa luffa seedlings in the three low-temperature treatment groups,there were 4945 up-regulated genes and 5065 down-regulated genes,involving 24 biological processes.Differentially expressed genes were involved in metabolic processes,plant hormone signal transduction,endocytosis and plant-pathogen interaction.In conclusion,low temperature and low light caused great damage to the photoprotection system of Luffa cylindrica seedlings,and the decrease of photosynthetic efficiency may indirectly cause seedling dehydration and wilting.SOD activity was the main physiological index of Luffa cylindrica seedlings responding to low temperature and low light stress.The differentially expressed genes are involved in biological pathways such as Ca2+ transport and trehalose metabolism.关键词
