不同化州柚品系新橘皮糖苷合成关键酶基因的差异表达分析
Differential Expression Analysis of Neohesperidin Synthesis Key Enzyme Genes of Citrus grandis'Tomentosa'from Different Strains

Abstract
Objective:To explore the differentially expressed genes of neohesperidin(NEO)synthesis key enzyme and its sequence variation of Citrus grandis'Tomentosa'from five strains.Methods:The traits of different strains of Citrus grandis'Tomentosa'were inves-tigated and analyzed.The NEO synthesis key enzyme genes differentially expressed in the strains were screened based on transcriptome data,and the expression levels were verified by RT-qPCR.The cloning and sequence alignment of candidate genes,protein property a-nalysis,evolutionary tree construction and other bioinformation analysis were conducted.Results:The characters of Miye and Miyehon-grou strains were highly similar,the expression levels of CgT1,2RhaT,the key enzyme genes for NEO synthesis,in the young fruits of the two strains were lower than those of other strains.The length of CgT1,2RhaT coding region was 1 359 bp,encoding 452 amino acids,which were highly homologous and intron-free,but there were multiple SNPs and InDels in the promoter region of this gene in Miye,Miyehongrou strains.CgT1,2RhaT were predicted to be unstable soluble non-secretory proteins,which were closely related to Cm1,2RhaT.Conclusion:The CgT1,2RhaT differentially expressed genes of neohesperidin synthesis key enzyme are cloned from five strains of Citrus grandis'Tomentosa'.It is found that the expression levels of CgT1,2RhaT genes from two strains of Citrus grandis'To-mentosa'with similar traits are lower,which may be related to the mutation site of their promoter region.The findings provide basic re-search for elucidating the molecular mechanism of the difference of component accumulation and functional gene differential expression in different strains of Citrus grandis'Tomentosa',and provide genetic resources for breeding of Citrus grandis'Tomentosa'varieties.关键词
