糖通饮靶向microRNA-21调控Smad7、PTEN抑制糖尿病肾病肾纤维化
Study on Tangtongyin Inhibiting Renal Fibrosis of Diabetic Kidney Disease by Targeting microRNA-21 to Regulate Smad7 and PTEN

Abstract
Objective:To explore the mechanism of Tangtongyin inhibiting renal fibrosis in diabetic nephropathy.Methods:Ten of the 60 SD rats were randomly selected as the normal control group,and the rest were fed with high sugar and high fat diet combined with multiple low dose intraperitoneal injection of STZ to establish a diabetic nephropathy(DKD)rat model.The successful DKD rats were randomly divided into model group,irbesartan(13.5 mg/kg)positive control group,the low(6.21 g/kg),medium(12.42 g/kg),high(24.84 g/kg)dose groups of Tangtongyin,each group of 10 rats was gavaged once a day for a total of 8 weeks.FBG and 24 hours urine mAlb of rats in each group were detected.The renal histopathological changes were observed by HE and Masson staining.The expres-sions of Smad7,PTEN,E-cadherin,FN,Vimentin and α-SMA proteins in rat kidney were detected by Western Blot.The expressions of Smad7,PTEN,FN,Vimentin α-SMA,E-cadherin mRNA and miRNA-21 were detected by RT-PCR.In vitro,HBZY-1 cells were trans-fected with miRNA-21 overexpression lentivirus and treated with Tangtongyin drug-containing serum.The expressions of Smad7,PTEN and α-SMA proteins were detected by Western Blot.The mRNA expressions of miR-21,Smad7,PTEN and α-SMA were detected by RT-PCR.Results:The in vivo experimental results showed that compared with the model group,Tangtongyin could significantly reduce the mRNA and protein expressions of FN,Vimentin and α-SMA in renal tissues of DKD rats,and significantly increase the mRNA and pro-tein expressions of Smad7,PTEN and E-cadherin.The expression of miRNA-21 and the levels of FBG and mAlb in urine at 24 hours were significantly decreased(P<0.05),and the renal histopathological changes of DKD rats were improved.The results of in vitro ex-periments showed that after transfection with miRNA-21 overexpressing lentivirus,the mRNA and protein expressions of Smad7 and PTEN in HBZY-1 cells in rats under the high sugar state were significantly decreased,and the mRNA and protein expressions of α-SMA were significantly increased.Tangtonguin could significantly decrease the miRNA-21 expression in HBZY-1 cells,the mRNA and protein expressions of Smad7 and PTEN were significantly increased,while the mRNA and protein expressions of α-SMA were significantly de-creased(P<0.05).Conclusion:miRNA-21 is one of the targets of Tangtongyin for prevention and treatment of DKD.Tangtongyin can positively regulate the expressions of its target Smad7 and PTEN by acting on miRNA-21,and inhibit renal fibrosis in diabetic nephropa-thy.关键词
