miR-130a/b基因家族进化分析及其在猪睾丸组织中的表达变化分析
Molecular Evolution Analysis of miR-130a/b Gene Family and Its Expression in Testicular Tissue of Pigs

Abstract
The purpose of this study was to investigate the evolution of miR-130a/b gene family and the expression of miR-130a/b in different development stages of porcine testis.miR-130a/b sequences available in the miRBase database were used to retrieve all species,and the structural location of miR-130a/b in the genome was further determined using the Ensembl database.The phylogenetic tree of miR-130a/b sequenece was constructed using Mega 10.0.The expression changes of miR-130a/b were detected by qRT-PCR at different developmental stages(D1,D30,D60,D90,D120,D150,D180)in testicular tissues of Shaziling pigs.The results of miRBase database showed that a total of 88 miR-130a/b sequences were retrieved,which was evenly distributed in 47 species,and they were composed of mature microRNA highly similar from different precursors of miR-130a/b,widely distributed and highly conserved.Most of miR-130a/b were located in the gene spacer region.According to phylogenetic tree analysis,the evolutionary direction of miR-130a/b gene family in all species had strong consistency.The results of qRT-PCR showed that the expression trend of miR-130a/b was the same in different developmental stages of the testicular tissue of Shaziling pigs.In other words,the expression level gradually increased during D1~D30 period,began to decrease during conservation period,and began to show a slow increase trend in sexual maturity period.The results of this study provided a certain reference for further exploration of the function and mechanism of miR-130a/b in porcine testicular tissue.关键词
