Molecular epidemiology investigation of porcine rotavirus in some regions of China in 2021-2022

为了解我国部分地区猪场猪轮状病毒(porcine rotavirus,PoRV)的流行情况及分子特征,对 2021-2022 年我国部分省份的 86 个规模化猪场的6 472 份腹泻样品进行检测,用RT-PCR方法调查PoRV的流行率,并对PoRV阳性样本进行VP7 和VP4 基因扩增、测序,使用MegAlign软件进行同源性分析,利用 MEGA11.0 构建系统进化树.结果显示,PoRV 的样品阳性率为 27.89%(1 805/6 472),猪场阳性率为 65.12%(56/86);从 98 份阳性PoRV样本中扩增出 76 个VP7 片段,G9 为优势基因型(42.11%),基因型G5、G26、G3、G4、G1、G2 和 G11 各占26.32%、9.21%、6.58%、6.58%、3.95%、2.63%和2.63%.扩增出的 35 个VP4 基因片段,以P[13]型为主,占 57.14%;其次为P[7]、P[23]、P[3]和P[6]型,分别占20%、14.29%、5.71%和2.86%.35 个毒株成功鉴定出G/P基因型,G9P[13](28.57%)为优势组合基因型,其他基因型为G5P[13](11.43%)、G4P[13](8.57%)、G9P[23](8.57%)、G26P[13](5.71%)、G1P[7](5.71%)、G26P[3](5.71%)、G5P[7](5.71%)、G3P[7](2.86%)、G11P[23](2.86%)、G5P[23](2.86%)、G11P[7](2.86%)、G3P[13](2.86%)、G5P[6](2.86%)和G4P[7](2.86%),其中G11P[7]为国内首次鉴定到的基因型.综上,目前PoRV的流行率高且基因型复杂,优势组合基因型为G9P[13].该结果丰富了PoRV的分子流行病学资料,对我国猪PoRV感染的防控具有重要意义,也为新型疫苗研发提供了参考依据.

In order to understand the prevalence and molecular characteristics of porcine rotavirus(PoRV)in some pig farms in China,in the period from 2021 to 2022,6 472 diarrhea samples were collected from 86 large-scale pig farms in some provinces of China and were studied.The prevalence of PoRV was investigated using RT-PCR,and the VP7 and VP4 genes in the PoRV positive samples were amplified and sequenced.Then,homology analysis was performed using the MegAlign software,and a phylogenetic tree was constructed using MEGA11.0.A total of 1 805 out of 6 472 diarrhea samples were positive for PoRV,with a positive rate of 27.89%(1 805/6 472)of all the samples and 65.12%(56/86)in the pig farms.76 VP7 fragments were amplified from 98 positive PoRV samples,G9(42.11%)was the dominant genotype,followed by G5,G26,G3,G4,G1,G2,and G11 which accounted for 26.32%,9.21%,6.58%,6.58%,3.95%,2.63%,and2.63%of the samples,respectively.The P[13]genotype(57.14%)was dominant among the 35 P types,followed by P[7],P[23],P[3],and P[6]types,accounting for 20%,14.29%,5.71%,and 2.86%,respectively.Furthermore,the G/P com-bination genotype was successfully identified in the 35 strains,and the dominant combination genotype was G9P[13](28.57%),the other weaker combinations were G5P[13](11.43%),G4P[13](8.57%),G9P[23](8.57%),G26P[13](5.71%),G1P[7](5.71%),G26P[3](5.71%),G5P[7](5.71%),G3P[7](2.86%),G11P[23](2.86%),G5P[23](2.86%),G11P[7](2.86%),G3P[13](2.86%),G5P[6](2.86%),and G4P[7](2.86%).Obviously,G11P[7]was first identified in China.Taken together,the current prevalence of PoRV was high and the genotypes were complex,with the dominant combina-tion genotype being G9P[13].The results of this study enriched the molecular epidemiology data of PoRV,which would be of great signifi-cance for the prevention and control of PoRV infection in pigs in China,and would also provide reference for the development of new vac-cines.
