Abstract

Objective To investigate the effect of TanshinoneⅡA on cognitive function in epileptic rats and its possible mo-lecular mechanism.Methods Clean grade SD rats were selected and randomly divided into control group,model group and low-,medium-and high-dose TanshinoneⅡA intervention group,with 10 rats in each group.Lithium chloride and pilocarpine were used to construct epileptic rat models.The control group was given normal saline intragastrically,and the model group was given saline intragastrically.Low-,medium-and high-dose intervention groups were given 5,10 and 20 mg/(kg·d)TanshinoneⅡA,respec-tively.The effects of TanshinoneⅡA on cognitive behavior of epileptic rats were detected by Morris water maze test.The expression levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)in brain tissues of each group were detected by ELISA.The expression levels of interleukin-1β precursor(pro-IL-1β)and NOD-like receptor protein 3(NLRP3)in brain tissues of each group were detected by Western blot.Results After intervention with TanshinoneⅡA for 6 d,compared with the control group,the es-cape latency of rats in model group was significantly prolonged,and the expressions of TNF-α and IL-1β and pro-IL-1β and NLRP3 in brain tissue were significantly increased(P<0.05).Compared with model group,the frequency of crossing the original platform and the residence time in the original platform were significantly increased or prolonged in medium-and high-dose intervention groups,while the expression levels of TNF-α and IL-1β in brain tissue were significantly decreased(P<0.05).Compared with model group,the protein expressions of pro-IL-1β and NLRP3 in brain tissue of rats in low-,medium-and high-dose intervention groups were significantly decreased(P<0.05),and gradually decreased with the increase of intervention dose of TanshinoneⅡA.Conclusion TanshinoneⅡA can significantly ameliorate cognitive dysfunction in epileptic rats,and ameliorate the degree of inflam-mation in brain tissue by regulating NLRP3/IL-1 βlevel,thereby playing a neuroprotective role.
