Effect of DA-6 and Coconut Juice on Strong seedling and Rooting of Tissue Culture Seedling of Anoectochilus roxburghii

Abstract
Objective:To scientifically address the issues of slenderness,low rooting coefficient,and low uniformity of tissue cul-tured seedlings in the rapid propagation technology of Anoectochilus roxburghii.The goal was to optimize the technology system of tissue culture and rapid propagation of Anoectochilus roxburghii and improve the quality of seedlings.Methods:Using N,N-diethyl aminoethyl hexanoate(DA-6)and coconut juice as experimental factors,the method of factorial analysis of variance designs was employed to inves-tigate the impact of 25 different combination treatments on the rooting ability of strong Anoectochilus roxburghii adventitious buds.Re-sults:5 to 10 mg/L DA-6 and 10% to 15% coconut juice had a better effect on plant height,and 5 to 15 mg/L DA-6 and 5% to 10% coconut juice promoted the growth of Anoectochilus roxburghii roots.The treatment group added with DA-6 and coconut juice at the same time had an obvious synergistic effect.The effect of strong seedling and rooting treated with 10 mg/L DA-6 and 10% to 15% coconut juice(T15,T16)was better than other single or combined treatment groups.Conclusion:Based on the comprehensive evaluation of in-dex values such as plant height,root quality,stem thickness,leaf number and fresh weight per plant,the best medium for promoting the strong seedling and rooting of the adventitious buds of Anoectochilus roxburghii is T15(MS with 4 mg/L IBA,0.5 mg/L NAA,2% su-crose,1% carrageenan,10 mg/L DA-6,10% coconut juice).The growth and development quality of tissue culture seedlings is better than the other 24 treatments.The research results provide a new scientific basis for the optimization and industrialization of the tissue culture and rapid propagation technology system of Anoectochilus roxburghii.关键词
