家鸡GBE1基因克隆与生物信息学分析
Cloning and Bioinformatics Analysis of GBE1 Gene in Gallus gallus

Abstract
Objective:To clone 1,4-α-glucan branchase 1(GBE1)gene and analyze its encoding protein bioinformatics and differ-ential expression.Methods:The full-length cDNA of GBE1 was amplified by PCR based on the transcriptome data of Gallus gallus and analyzed by bioinformatics.qRT-PCR assay was used to detect gene expression differences in 6 tissues of domestic Gallus gallus.Re-sults:The full-length of GBE1 gene was 2 145 bp and the open reading frame(ORF)was 2 130 bp,encoding 709 amino acids.The rela-tive molecular weight of the GBE1 protein was 81.993,the theoretical isoelectric point was 6.25,the stability index was 35.40,and the average hydrophilic value was-0.484,which belonged to stable acidic hydrophilic protein.GBE1 protein contained 67 phosphorylation sites,and there was no transmembrane region and signal peptide sequence.It was mainly located in cytoplasm,nucleus,mitochondria and cellular fluid.The secondary structure mainly consisted of α-helix,β-folding,random curling and extension chains,and the tertiary structure showed that the protein was a monomer.The homology analysis showed that the amino acid sequence of GBE1 protein in do-mestic Gallus gallus was closely related to that of Numida meleagris.The qRT-PCR results showed that GBE1 gene was expressed in all the tissues of heart,liver,spleen,lung,kidney and endothelium corneum gigeriae galli,but the expression level in liver and endothelium corneum gigeriae galli was significantly higher than that in other organs.Conclusion:The results of this study lay a foundation for further research on the function of GBE1 gene and the regulation mechanism of dextran branch enzyme in Gallus gallus.关键词
