摘要

Abstract

Objective:To explore the differences of coumarin content and IPT gene relative expression in Glehniae Radix,so as to provide theoretical basis for quality assessment and further selection and breeding of Glehniae Radix.Methods:HPLC method was used to determine the contents of 9 coumarins in Glehniae Radix.The relative expression of isopentenyltransferase gene(IPT)was detected by real-time quantitative PCR.Results:The contents of coumarin in Glehniae Radix cultivated in Laiyang,Chifeng and wild species in Baxiandao,Jinshitan and Haiyang were higher.The relative expression of IPT gene was higher in Glehniae Radix from Chifeng and Haiyang.IPT gene expression was significantly positive correlated with psoralen and isophellolactone contents(P<0.01).Principal com-ponent analysis showed that Glehniae Radix cultivated in Laiyang and Haiyang were the planting area of high quality Glehniae Radix.Conclusion:In this study,the differences of coumarin content and IPT gene expression in Glehniae Radix from 8 provenances and the correlation between them are investigated,which can provide reference for the selection of provenances with high application value,and contribute to the further development and utilization of Glehniae Radix.
