苗药九仙罗汉接骨汤含药血清对MC3T3-E1Subclone14细胞增殖、凋亡和Runx2、Osterix、Bax、Bcl-2表达的影响
Study on the Effects of Miao Medicine Jiuxian Luohan Jiegu Decoction Drug Serum on the Proliferation,Apoptosis and Runx2,Osterix,Bax,Bcl-2 Expressions of MC3T3-E1Subclone14 Cells

Abstract
Objective:To observe the effects of Jiuxian luohan jiegu decoction drug serum on the proliferation and apoptosis of MC3T3-E1Subclone14 cells and the expressions of Runx2,Osterix,Bax,Bcl-2 mRNA and proteins,to explore the possible mechanism of promoting the proliferation and inhibiting the apoptosis of osteoblasts.Methods:The MC3T3-E1Subclone14 cell line cultured in vitro was divided into blank control group,Xianling gubao capsule group,Jiuxian luohan jiegu decoction drug serum low,medium and high doses groups.After preparation of drug serum,all cells in each group were cultured with high glucose DMEM medium with different concentra-tions.Cell proliferation and apoptosis were detected by MTT and flow cytometry after 24 hours.The mRNA and protein expressions of Runx2,Osterix,Bax and Bcl-2 were determined by RT-PCR,Western Blot and immunohistochemistry.Results:Compared with blank control group,the multiplication capacity of cells in Jiuxian luohan jiegu decoction medium and high doses groups was significantly in-creased,the apoptosis rate was significantly decreased,the protein expressions of Runx2,Osterix and Bcl-2 mRNA were significantly in-creased,and the mRNA and protein expression of Bax was significantly decreased(P<0.05 or P<0.01).Conclusion:Miao medicine Ji-uxian luohan jiegu decoction can promote the proliferation and inhibit the apoptosis of MC3T3-E1Subclone14 cells,and its mechanism may be related to the regulation of Runx2,Osterix,Bax,Bcl-2 genes and protein expressions.关键词
