山茱萸的化学成分及其抗阿尔茨海默病作用研究
Study on the Chemical Constituents of Cornus officinalis and Anti-Alzheimer's Disease Effect

Abstract
Objective:To study the chemical constituents and anti-Alzheimer's disease effect of Cornus officinalis.Methods:Macro-porous adsorption resin,silica gel,MCI,Toyopearl HW-40C,semi-preparative HPLC and other chromatographic methods were used to i-solate and purify the compounds of the aqueous extract of Cornus officinalis.The structures were identified by NMR spectroscopy,mass spectrometry and other spectroscopic techniques.The anti-Alzheimer's disease effect of the compounds was evaluated in vitro using PC-12 cells injury induced by Aβ25-35.Results:Twelve compounds were isolated from the aqueow extract of Cornus officinalis and identified as quercetin-3-O-β-D-glucuronide-6″-ethyl ester(1),kaempferol-7-O-β-D-glucoside(2),2α,3β-dihydromyricetin(3),naringenin-7-O-β-(6″-galloylglucoside)(4),6′-O-vanilloyltachioside(5),dunnianoside D(6),3′,5′-di-C-β-glucopyranosylphloretin(7),4-methoxy-benzoic acid(8),p-hydroxybenzoic acid ethyl ester(9),p-hydroxyl-benzyl-propanol(10),protocatechuic acid(11),gallic acid-3-methyl ether(12),respectively.In vitro activity screening results indicated that compounds 3,4,9 and 12 could effectively alleviate the damage of PC-12 cells induced by Aβ25-35.Conclusion:Among them,compounds 1 to 5 and 7 to 10 are isolated from Cornus officinalis for the first time.Compounds 3,4,9 and 12 show potential anti-Alzheimer's disease effect.关键词
