摘要

Abstract

Objective:To investigate the effects and mechanism of Zicui decoction on glucose and lipid metabolism in the liver of type 2 diabetic rats.Methods:10 SD rats were randomly selected as the normal control group from 90 SD rats.The other rats were given a high sugar and high-fat diet for 4 weeks as well intraperitoneally injected a low dose of streptozotocin(STZ,35 mg/kg)to replicate the type 2 diabetic rat model.The successfully modeled rats were randomly divided into Zicui decoction high-(15.4 g/kg),medium-(7.7 g/kg),low-(3.85 g/kg)dose groups,metformin group(0.2 g/kg)and model group.Each group was administered once a day for 14 consecutive days by intragastric.The amount of diet and drinking water in each group were recorded;The liver was weighed and the liver index was calculated;The levels of AST,ALT,SOD,MDA and GSH in serum were measured;The pathological changes of liver were ob-served by HE staining;Hepatic lipid deposition was observed by liver oil red O staining;The hepatocyte glycogen particle area was ob-served by PAS staining.The protein expressions of p-AMPK,p-ACC1,p-SERBP-1,p-PPARα in liver tissue were measured by Western Blot.Results:Compared with normal control group,food intake,water intake and liver index of model group were significantly increased(P<0.05 or P<0.01),liver cell morphology was significantly changed,lipid deposition area and serum ALT,AST and MDA levels were significantly increased.The levels of serum SOD and GSH,hepatocyte glycogen particle area,the protein expressions of p-AMPK,p-ACC1,p-SERBP-1 and p-PPARα in liver tissue were significantly decreased(P<0.01).Compared with model group,Zicui decoction could significantly reduce diet,water intake and liver index of diabetic rats(P<0.05 or P<0.01),improve liver tissue injury,and signif-icantly reduce lipid deposition area in liver tissue and serum AST,ALT and MDA levels.The levels of SOD and GSH in serum,hepato-cyte glycogen particle area,the protein expression of p-AMPK,p-ACC1,p-SERBP-1 and p-PPARα in liver tissue were significantly in-creased(P<0.05 or P<0.01).Conclusion:Zicui decoction can regulate liver lipid metabolism,the mechanism may be related to acti-vating AMPK-PPARα signaling pathway,promoting lipid metabolism,reducing oxidative stress level,improving antioxidant capacity.
