Study on the Quality of Sargassum fusiforme Based on PMP Pre-column Derivatization Reaction Combined with Multivariate Chemometrics and QAMS

Abstract
Objective:To establish HPLC fingerprint and QAMS method for distinguishing Sargassum fusiforme decoction pieces from formula granules,and to screen key difference marker compounds,so as to provide reference for improving quality control standards of Sargassum fusiforme.Methods;According to the principle of precolumn derivatization of carbohydrate compounds,HPLC was used to determine the fingerprints of Sargassum fusiforme decoction pieces and formula granules.Systematic cluster analysis(HCA),principal component analysis(PC A)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to screen the difference markers.Using fucose as reference object,QAMS method was established for the determination of main monosaccharides in Sargassum fusiforme decoction pieces and formula granules.Results:The established fingerprint and QAMS method could effectively distinguish the decoction pieces and formula granules of Sargassum fusiforme.Fucose,D-galactose,D-glucose and two unknown compounds(peaks 5 and 10)were used as difference markers.However,the contents of D-mannose and fucose in formula granules were significantly differ-ent from those in decoction pieces.Conclusion:The fingerprint and four monosaccharide content determination method established in this study is stable and reliable,which can be used for quality control of Sargassum fusiforme decoction pieces and formula granules.关键词
