目的:通过转录组学测序技术(RNA-Seq)探究经穴推拿对产后大鼠基因差异性表达的影响,并分析生物机制特征.方法:采用随机数字表法将6只健康雌性大鼠随机分为空白对照组、经穴推拿组,每组3只,与雄性大鼠合笼交配.经穴推拿组大鼠产后立刻连续推拿干预7天.取大鼠腹主动脉血提取RNA,采用RNA-Seq技术进行转录组学测序,将筛选的差异基因进行GO和KEGG生物信息学分析.结果:经穴推拿组与空白对照组大鼠相比,共959个差异表达基因,其中889个表达上调,70个表达下调.GO分析发现差异基因主要集中在代谢、合成、免疫相关生物学过程,KEGG分析发现差异基因主要参与信号转导、免疫系统、运输和分解通路,并与细胞的生长、分化、衰老与凋亡相关.结论:经穴推拿能够影响产后大鼠的基因表达水平,参与调控产后机体代谢、免疫、细胞衰老进程,为推拿干预产后机能恢复提供了研究基础,有助于未来进一步挖掘经穴推拿促进产后恢复的作用机制.

Objective:To investigate the effect of tuina on differential expression genes in postpartum rats by RNA sequencing(RNA-Seq)and ana-lyze the characteristics of biological mechanism.Methods:6 female SD rats were randomly divided into Control Pregnancy Group(CP)and Tuina Pregnancy Group(TP),with 3 in each group,then mated with male SD rats.Rats of TP were gived Tuina intervention for 7 days.RNA-Seq technique was used to detect the differential expression genes of blood,Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis were performed for all rats.Results:Compared to CP,there were 959 differential expression genes in TP.GO analysis showed that the genes were mainly involved in metabolic,synthetic,and immune biological processes.KEGG analysis showed that the genes were primarily associated with the pathways of signal transduction,immune system,transport and catabolism,which related cell growth,differentiation,senescence,and apoptosis.Con-clusion:Tuina could regulate the gene expression in postpartum rats to participate in metabolism,immunity,and cell senescence,which provides evi-dence and support for Tuina adopting in postpartum functional recovery.
