Abstract

[Objective]Analyzing the genetic diversity and building a fingerprint map of sweetpotato varieties based on morphological indicators and molecular markers provide theoretical reference for the study of parent selection and ge-netic background of sweetpotato breeding in Fujian province.[Method]28 pairs of ISSR primer,27 pairs of SRAP prim-ers and basic agronomic traits were screened for genetic diversity analysis of 24 sweetpotato cultivars.SPSS 26 was used to perform clustering analysis based on agronomic traits using the intergroup connection method and the mean European distance method.Observed number of alleles(Na),Effective number of alleles(Ne),Shannon's information index(I)and Nei's gene diversity(H')were performed by Popgene 32.NTSYS pc 2.1 was used for the genetic similarity coeffi-cients and the clustering analysis based on molecular markers.The fingerprint was constructed according to the primer amplification results.[Result]The results showed that 28 pairs of ISSR primers and 27 pairs of SRAP primers amplified 362 and 364 bands,respectively,with an average polymorphism ratio of 78.73%and 83.791%,and the polymorphism rate of some primers was as high as 100%.24 germplasms were divided into three categories at the Euclidean distance of 8 by morphological cluster analysis.The first category was Quanshu 76,Quanshu 12 and Longshu 14,and the second category included Xuzishu 2,Quanshu 10,Pushu 20,Fushu 24,Puzishu 18 and Funingzi 3,and the rest were in the third category.The polymorphism detection efficiency of ISSR and SRAP primers was high.The genetic similarity coefficient was between 0.674-0.851,the average value was 0.737,and when the genetic similarity coefficient was 0.728,it could be divided into three categories,Funingzi 3 was the first category,Fushu 24,Xuzishu 2,Pushu 20,Pushu 53,Puzishu18,Puzi-shu 3,Fushu 404 and Pushu 16 were the second category,and the rest were divided into the third category based on ISSR markers.Based on SRAP marker analysis,the genetic similarity coefficient was between 0.591-0.879,the average value was 0.712,and these materials were divided into 3 categories,Quanshu 76 was the first category,Puzishu 18 and Xuzishu 2 were the second category,and the rest of the materials were divided into the third category at the genetic similarity coef-ficient of 0.696.The primer UBC899 was used to construct the fingerprint map of 24 sweetpotato resources.[Conclusion]Both ISSR and SRAP molecular markers are suitable for sweetpotato material analysis,the genetic diversity of the 24 sweetpotato materials is relatively rich,and the ISSR molecular clustering results are more consistent with the pedigree of the germplasm resources,and the primers UBC899 can be used to distinguish the tested sweetpotato varieties.
