康复新液对糖尿病难愈合创面中AGEs、RAGE及HIF-1表达水平的影响
Investigating changes in AGEs,RAGE,and HIF-1 expression levels in diabetes non-healing wounds with the intervention of Kangfuxin Solution

Abstract
Objective To apply Kangfuxin Solution in the intervention of non-healing wounds in diabetic pa-tients,to observe changes in the expression levels of advanced glycation end products(AGEs),receptor for advanced gly-cation end products(RAGE),and hypoxia-inducible factor-1(HIF-1),and to preliminarily explore the correlation between the AGEs/RAGE pathway and HIF-1 in non-healing wounds of diabetes.Methods Ten cases of diabetic foot patients admitted to Beilun People's Hospital of Ningbo,from January to December 2020,were selected for the study.The healing conditions of the wounds were observed at 0,7,14,and 28 days of medication.ELISA was used to detect changes in hydroxyproline content in granulation tissue at different medication times.Human dermal microvascular endo-thelial cells(HMVECs)were cultured in vitro and randomly divided into control group(conventional culture),AGE-HSA group(220 nmol/L AGE-HSA),and Pidotimod+AGE-HSA group(220 nmol/L Pidotimod+AGE-HSA).Western blot was used to detect the expressions of AGEs,RAGE,and HIF-1α proteins in wound tissue and HMVECs.Results After 7,14,and 28 days of medication,the wounds of diabetic patients healed well,with an increasing trend in the effective rate of drug treatment and hydroxyproline content.The expression levels of AGEs,RAGE,and HIF-1α pro-teins showed a decreasing trend.Compared with 0 day,the hydroxyproline content in the granulation tissue of the wounds significantly increased at 7,14,and 28 days(P<0.01),while the expression levels of AGEs,RAGE,and HIF-1αproteins were significantly downregulated(P<0.05 or P<0.01).Western blot results of HMVECs showed that compared with the control group,the expression of AGEs,RAGE,and HIF-1 α proteins in the AGE-HSA group were significantly upregulated(P<0.01),and Pidotimod significantly inhibited this upregulation(P<0.05 or P<0.01).Conclusion The prognosis of diabetes non-healing wounds is closely related to the expression levels of AGEs,RAGE,and HIF-1α.The expression of HIF-1α may be associated with the activation of the AGEs/RAGE signaling pathway.关键词
