GPR176 mRNA与胃癌预后及免疫治疗反应关系的生物信息学分析
Bioinformatics analysis on the relationship of GPR176 mRNA with prog-nosis of gastric cancer and response to immunotherapy

Abstract
Aim:To investigate the expression level of G protein coupled-receptor(GPR176)in gastric cancer by bioinformatics,and explore its potential in predicting the prognosis and immune checkpoint blocker response in patients with gastric cancer.Methods:R software was used to analyze the expression difference of GPR176 mRNA in gastric cancer and normal gastric tissue using TCGA-STAD data set and GEO data set,and immunohistochemical image of Human Protein Atlas was used to analyze the expression of GPR176 protein in gastric cancer and normal gastric tissue.The expression of GPR176 mRNA in gastric cancer patients with different clinical features was compared using TCGA-STAD data set.The value of GPR176 mRNA in predicting prognosis of gastric cancer and immune checkpoint blocker response was explored using the TCGA-STAD data set and GEO data set.Results:Both mRNA and protein expressions of GPR176 were significantly up-reg-ulated in gastric cancer tissue,which was associated with adverse clinical features of patients with gastric cancer.Patients with higher GPR176 mRNA had shorter overall survival.ROC curve analysis showed that GPR176 mRNA was of some value in predicting the 1,3,5 year survival of gastric cancer.The gastric cancer patients with higher GPR176 mRNA had higher infiltration abundance in resting NK cells and macrophages M2,and lower immunophenotypic scores.Conclusion:GPR176 is highly expressed in gastric cancer tissue.It could be used as a potential novel molecular marker for predicting prognosis of gastric cancer and immune checkpoint blocker response.关键词
