Abstract
Objective:To investigate the effect and mechanism of resveratrol on diabetic cardiac microvascular injury.Methods:Rat cardiac microvascular endothelial cells were isolated and cultured,and randomly divided into normal glucose group,mannitol group,high glucose group,high glucose + resveratrol group and high glucose + resveratrol + compound C group.The cellular reactive oxygen species level was detected by flow cytometry,superoxide dismutase activity and malondialdehyde content were detected by commercial assay kit,and the activity of AMP-activated protein kinase(AMPK)was detected by Western blot.Results:Compared with the normal group,reactive oxygen species level and malondialdehyde content were increased,and superoxide dismutase activity and p-AMPK/AMPK value were decreased in the high glucose group.Compared with the high glucose group,reactive oxygen species level and malondialdehyde content were decreased,and superoxide dismutase activity and p-AMPK/AMPK value were increased in the high glucose + resveratrol group.Compared with the high glucose + resveratrol group,reactive oxygen species levels and malondialdehyde content were increased and superoxide dismutase activity and p-AMPK/AMPK values were decreased in the high glucose + resveratrol + compound C(AMPK inhibitor)group,and the differences were statistically significant(all P<0.05).Conclusion:Resveratrol attenuates diabetic cardiac microvascular injury by activating AMPK.关键词
