Effect of BMP-2/Smads/Osterix signal on proliferation and differentiation of rat osteoblasts induced by sodium fluoride

Abstract
Objective:To investigate the effect of sodium fluoride on the proliferation and differentiation of cranial osteoblasts in rats,and to further study the regulatory mechanism of BMP-2/Smads/Osterix signaling.Methods:Osteoblasts were i-solated from the skull of neonatal SD rats(within 24 h)and cultured in vitro.The osteoblasts were treated with different concentrations of sodium fluoride,and the effects of sodium fluoride on osteoblasts were observed with MTT and alkaline phosphatase.The expressions of BMP-2 protein,Smad1/5/9 protein and Osterix protein were determined by western blot.Results:Compared with the control group,osteoblasts proliferate significantly and alkaline phosphatase activity is en-hanced when exposed to low doses of sodium fluoride(0.25 mmol/L,0.50 mmol/L),while osteoblasts are stimulated by high doses of sodium fluoride(2.00 mmol/L,4.00 mmol/L).The proliferation of osteoblasts was significantly inhibited and the activity of alkaline phosphatase was decreased.Compared with the control group,the expression levels of BMP-2 protein,Smad1/5/9 protein and Osterix protein were significantly increased when the concentration of low dose sodium fluoride was 0.50 mmol/L,with statistical significance(P<0.05).Conclusion:Low concentration sodium fluoride can promote the proliferation and differentiation of osteoblasts through BMP-2/Smads/Osterix signal,while high concentra-tion sodium fluoride can inhibit the proliferation and differentiation of osteoblasts.关键词
