青蒿素-丙酮层共组装纳米药制备、表征及体外抗肿瘤初步评价OACSTPCD

为解决青蒿素及其丙酮层(以多甲氧基黄酮为主成分的青蒿素工业提取废料)的水溶性差导致其抗肿瘤应用受限的问题,采用乳化-溶液挥发法,构建质量均一、结构稳定的青蒿素-丙酮层超分子共组装纳米药.应用激光粒度仪、傅里叶变换红外光谱仪及扫描电子显微镜等对其进行表征,并以MTT、划痕试验测定纳米药对小鼠源CT26结肠癌细胞增殖、迁移能力的影响.结果表明:所制备的纳米药平均粒径106.93 nm,载药量7.78%,包封率72%;MTT试验检测共组装药物(青蒿素、丙酮层量之比1∶2)IC5.为青蒿素、丙酮层浓度比17.13 mg·L-1∶34.27 mg·L-1,显著优于各药单用组;划痕试验表明共组装药物对肿瘤细胞迁移具有显著抑制作用.综上,制备的青蒿素-丙酮层超分子共组装纳米药具有较强抗肿瘤活性,为抗肿瘤药物开发和中药提取废弃物循环利用奠定了重要基础.

To solve the problem of poor water solubility of artemisinin and its acetone layer(industrial extraction waste of artemisinin with polymethoxyflavonoids as the main component)which leads to the limitation of its antitumor applica-tion,the supramolecular co-assembled nanomedicine of artemisinin and acetone layer with homogeneous quality and stable structure was constructed by using the method of emulsification-solution volatilisation.Laser particle size apparatus,fou-rier transform infrared spectrometer and scanning electron microscope were applied to characterize them,and MTT and scratch assay were used to determine the effects of nanomedicines on the proliferation and migration ability of mouse-de-rived CT26 colon cancer cells.The results showed that the average particle size of the prepared nanomedicine was 106.93 nm,the drug loading capacity was 7.78%,and the encapsulation rate was 72%;the IC50 of the co-assembled drug(artemisinin∶acetone layer=1∶2)was artemisinin∶acetone layer=17.13 mg·L-1∶34.27 mg·L-1,which was significantly better than that of the single-use group of the drugs,and the scratching experiment demonstrated that the co-assembled drug had a significant inhibition effect on the migration of the tumor cells.In conclusion,the supramolecular co-assembled nanomedicine of artemisinin-acetone layer prepared in this study has strong anti-tumor activity,which lays an important foundation for the development of anti-tumor drugs and the recycling of traditional Chinese medicine extract waste.
