HPLC法测定五味子颗粒中7种木脂素成分的含量OACSTPCD
Measurement of seven lignans in Schisandrae Chinensis Fructus Granules by HPLC
建立了同时测定五味子颗粒中7种木脂素成分含量的高效液相色谱(HPLC)法.采用Poroshell 120 SB-C18色谱柱(150 mm×4.6 mm,2.7 μm)进行分离,四氢呋喃水-乙腈梯度洗脱,流速为0.6 mL/min,柱温为30℃,检测波长为220 nm,进样量为10 μL.结果表明:该方法对五味子醇甲、五味子醇乙、五味子酯甲、五味子酯乙、五味子甲素、五味子乙素和五味子丙素的检测下限(LOD)分别为0.017、0.041、0.156、0.165、0.136、0.166和0.113 µg/mL,线性关系良好,相关系数(r)均大于0.999 9,平均加样回收率为98.1%~100.9%,相对标准偏差(RSD)均小于3.0%.因此,该方法重复性好、灵敏度高,所得含量关系可对五味子颗粒进行质量控制,特别是可为预防南五味子掺伪提供参考,也为其他含五味子制剂的质量标准修订奠定了基础.
A reliable HPLC method was established to measure the contents of 7 lignans in Schisandrae Chinensis Fructus Granules.The chromatographic process was performed on a Poroshell 120 SB-C18 column(150 mm×4.6 mm,2.7 μm)using gradient elution with the mobile phase of water-containing tetrahydrofuran and acetonitrile.With the optimized separation conditions(flow rate:0.6 mL/min;column temperature:30℃;detection wavelength:220 nm;injection volume:10 μL),the LODs for schizandrin,Gomisin A,Schisantherin A,Schisantherin B,Schisandrin A,Schisandrin B and Schisandrin C were found to be 0.017,0.041,0.156,0.165,0.136,0.166 and 0.113 μg/mL,respectively.Excellent linearity(r>0.999 9)was achieved for 7 studied lignans,and the average extraction recoveries(RSD<3.0%)ranged from 98.1%to 100.9%.Overall,our HPLC method demonstrated good reproducibility and high sensitivity,and provided an excellent tool in controlling the quality of Schisandrae Chinensis Fructus Granules,especially in identifying the harmful addition of Schisandrae Sphenantherae Fructus.In addition,this quantitative analysis approach laid a foundation for the revision of quality standards with regards to other products containing Schisandrae Sphenantherae Fructus.
