Investigation of the causal relationship between time spent on computer and myopia based on a Mendelian randomization

Abstract
Objective The two-sample Mendelian randomization method(MR)was used to investigate the causal relationship between time spent on computer and myopia.Methods The pooled data from gene-wide association studies(GWAS)were analysed.Genetic loci closely associated with myopia were collected as instrumental variables.Two-sample Mendelian randomization was performed by inverse variance weighting method,weighted median and Mr-Egger,respectively.Inverse variance weighting method,Mr-Egger and leave-one-out were used to test heterogeneity and sensitivity.Mr-Egger was used for pleiotropic analysis.Results The Inverse variance weighting method showed significant positive correlation between time spent on computer and myopia,with the odds ratio of 1.041[95% confidence interval,CI):1.023~1.060;P=5.69e-06].Weighted median and Mr-Egger showed similar causality.The result of Mr-Egger indicated the absence of pleiotropy.Conclusions There is a positive relationship between time spent on computer and myopia.关键词
