摘要

Abstract

In order to reveal the effect of cellular autophagy-related protein 9A(ATG9A)on foot-and-mouth disease virus(FMDV)replication,the co-localization of FMDV and ATG9A as well as the im-pact on ATG9A expression during viral infection were detected by indirect immunofluorescence assay,reverse transcription quantitative real-time PCR and Western-blot methods.The recombinant plasmid for expression of ATG9A and small interfering RNA targeting ATG9A were constructed and employed to esti-mate the influence for adsorption,internalization,genomic replication,polyprotein translation,and progeny production of FMDV,respectively.The results showed that the FMDV was co-localized with ATG9A and the expression of ATG9A was down-regulated after FMDV infection.ATG9A could not affect virus ad-sorption,but significantly inhibited FMDV internalization.Furthermore,overexpress ion(or knock-down)of ATG9A dramatically reduced(or increased)FMDV proliferation and the production of progeny virions.In conclusion,ATG9A might be deleterious to FMDV replication,which would provide insights into the replication cycle of FMDV in susceptible cells.
