Polymorphic Multiple-base Repeat Microsatellite Loci of Chinese Tiger Frog Hoplobatrachus chinensis

Abstract
[Objective]Screening of high-quality polymorphism microsatellite markers in the Chinese tiger frog Hoplobatrachus chinensis can be used for identification,genetic diversity,population structure,and genetic linkage map analysis of this species.[Method]In this study,we identified 76 multiple-base repeat microsatellite loci,including 33 trinucleotide,42 tetranucleotide and 1 pentanucleotide repeat microsatel-lite loci from partial genome of Chinese tiger frog Hoplobatrachus chinensis using the next-generation sequencing technology.We genotyped 30 individuals from a population in an artificial pond at the herpetological laboratory of Lishui University,Zhejiang Province,China.[Result]The results showed that the mean number of observed alleles and polymorphic information content(PIC)per locus was 4(range from 2 to 7)and 0.514(range from 0.164 to 0.822),respectively.The observed and expected heterozygosity values ranged from 0.00 to 0.97 and from 0.18 to 0.82,respectively.Two loci(THC141 and THC377)showed significant departure from Hardy-Weinberg equilibrium and significant linkage disequilibrium was found among 11 pairwise loci.[Conclusion]These novel polymorphic microsatellite markers are expected to be val-uable for future studies on genetic diversity and conducting genetic breeding,enabling the formulation of effective conservation strategies and the use of molecular assisted selection for H.chinensis.关键词
