摘要

Abstract

In order to optimize and establish the tissue culture regeneration system of sycamore,this study took healthy and pest free annual stem segments as the explants,and investigated the effects of different disinfection methods and hormone ratios on disinfection,initiation,proliferation and rooting of explants.The results showed that the optimal disinfection method for the stem explants of sycamore was 75%ethanol disinfection for 30 s+0.1%HgCl2 disinfection for 17 min,and the contamination rate could reduce to 20%.The most suitable medium for initiation culture was MS+ZT 4 mg/L+IAA 1.0 mg/L,and the initiation rate could reach 65%.The most suitable proliferation medium was MS+ZT 3 mg/L+IAA 1.0 mg/L,and the proliferation coefficient could reach 1.95.The most suitable rooting medium was 1/2 MS+ IBA 0.1 mg/L,and the rooting rate could reach 75%.
