目的 研究菊茎叶总黄酮指纹图谱与其抗氧化活性的相关性,探究菊茎叶总黄酮抗氧化活性的物质基础.方法 采用高效液相色谱法建立 11 批菊茎叶总黄酮样品的指纹图谱;分别采用 1,1-二苯基-2-三硝基苯肼自由基清除法、2,2′-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基清除法及血浆铁离子还原法测定其抗氧化活性,结合灰色关联度法、偏最小二乘回归法分析谱效关系.结果 建立 17 个共有峰的菊茎叶总黄酮的高效液相色谱指纹图谱,指认其中 11 个共有峰,相似度大于 0.886.灰色关联分析显示其中 11 个色谱峰关联度均大于 0.9,偏最小二乘回归分析结果显示 6 个色谱峰的变量重要性投影值>1,表明菊茎叶总黄酮与抗氧化活性有重要相关性.结论 共有峰 6(山柰酚-3-O-芸香糖苷)、峰 11、峰 12(香叶木素-7-O-β-D-葡萄糖苷)、峰 13(木犀草素)、峰 15 与抗氧化活性密切相关,初步推测为菊茎叶总黄酮抗氧化活性的物质基础.

Objective To investigate the fingerprint of total flavonoids in chrysanthemum stems and leaves and its relationship with antioxidant activities,and to explore the material basis of the antioxidant activities of total flavonoids in chrysanthemum stems and leaves.Methods High-performance liquid chromatography(HPLC)was used to establish the fingerprints of 11 bat-ches of total flavonoids from chrysanthemum stems and leaves,and 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging method,2,2′-azinobis(3-ethylbenzthiazoline-6-sulfonate)(ABTS)radical scavenging method,and ferric reducing ability of plasma(FRAP)method were used to measure their antioxidant activities;spectrum-effect relationship was analyzed using grey correlation degree and partial least squares regression.Results The HPLC fingerprints of total flavonoids in chrysanthemum stems and leaves were established with 17 common peaks,among which 11 common peaks were identified with a similarity of greater than 0.886.The grey correlation analysis showed a correlation degree of>0.9 for all 11 chromatographic peaks,and the partial least squares regression analysis showed a variable importance projection value of>1 for 6 chromatographic peaks,sug-gesting a significant correlation with antioxidant activities.Conclusion The common peak 6(kaempferol-3-O-rutinoside),peak 11,peak 12(diosmetin-7-O-β-D-glucopyranoside),peak 13(luteolin),and peak 15 are closely associated with antioxidant activi-ties and are speculated to be the material basis of antioxidant activities of total flavonoids from chrysanthemum stems and leaves.
