Abstract
Objective:Endometrial carcinoma(EC)is a common malignant tumor of the female reproductive system,with endometrioid adenocarcinoma(EEC)being the most common subtype.The etiology of EEC remains unclear,although studies have shown that the expression of circular RNA(circRNA)is related to the progression of EEC.This study aims to detect the expression level of hsa_circ_0007067 in EEC,and explore its relationship with the clinicopathological features of EEC.
Methods:High-throughput sequencing analysis was used to analyze the gene expression levels of 2 EEC tissues and 2 normal endometrial tissues.Based on the criteria of |log2(Fold changes)|≥1.5 and P＜0.05,the most significant down-regulation of hsa_circ_0007067 was selected for further study.Real-time PCR was used to detect the relative expression levels of hsa_circ_0007067 in 36 EEC tissues(a EEC group)and 36 normal endometrial tissues(a control group).Bioinformatics analysis was used to predict the binding sites and coding capabilities of hsa_circ_0007067.
Results:The expression level of hsa_circ_0007067 in the EEC group was significantly lower than that in the control group(P＜0.05),and its expression level was related to Inter-national Federation of Gynecology and Obstetrics(FIGO)stage and histological differentiation.The receiver operating characteristic(ROC)curve showed an area under curve(AUC)of 0.936,with a sensitivity of 0.833 and specificity of 0.889,and the optimal cut-off value was 0.722,indicating statistically significant differences(P＜0.05).Hsa_circ_0007067 exhibited the potential to translate into peptides,with circbank database predicting a total of 24 potential mRNA binding sites.
Conclusion:Hsa_circ_0007067 is associated with the occurrence and development of EEC diseases,and may be served as a molecular marker for early screening and prognosis assessment of EEC.It may affect the progression of EEC diseases through its function of binding to downstream target gene miRNAs or encoding protein functions.关键词
