替罗非班对急性缺血性卒中小鼠的神经保护作用
Neuroprotective effects of tirofiban in mice with acute ischemic stroke

Abstract
Objective To investigate the effects of tirofiban on brain infarct volume and neuronal apoptosis in mice with acute ischemic stroke(AIS).Methods Sixty-five C57BL/6 mice were randomly divided into sham group,model group,and low-dose,medium-dose,and high-dose tirofiban groups,with 13 mice in each group.Amouse model of cerebral ischemia was es-tablished through photochemical thrombosis.The model group received injection of the same volume of normal saline via the tail vein,while the tirofiban groups receivedinjection of different doses of tirofiban into the tail vein.The neurological deficits of mice were evaluated using the modified neurological severity scale(mNSS).The brain infarct volume was measuredwith 2,3,5-triphe-nyltetrazolium chloride staining.Neuronal apoptosis was determined with TUNEL staining and NeuN staining.Results There were significant differences in the mNSS score,cerebral infarction volume,the number of TUNEL-positive cells,and the number of NeuN-positive cells between the five groups(F=9.630-124.927,P<0.05).Compared with the model group,the medium-dose tirofiban group showed significantly resolved neurobehavioral damage,a significantly decreased cerebral infarction volume,a signi-ficantly decreased number of TUNEL-positive cells,and a significantly increased number of NeuN-positive cells(P<0.05).Con-clusion A medium dose of tirofiban can play a neuroprotective role in mice with AIS,decreasing neurological deficit score,infarct volume,and neuronal apoptosis.关键词
