Abstract

Objective To investigate the expression and clinical significance of ASPM and KIF20A in esophageal squa-mous cell carcinoma(ESCC).Methods Based on GEO database,core genes were screened and their expression in ESCC were predicted.KEGG pathway was enriched to analyse potential signaling pathways involved in ASPM and KIF20A.TIMER database was used to analyse correlation between ASPM,KIF20A and infiltration of immunocells.95 ESCC tissues and 30 paraneoplastic normal tissues were selected,and expression of ASPM and KIF20A in ESCC and paraneoplastic normal tissues were detected by im-munohistochemistry and immunochemistry.Immunohistochemistry was used to detect protein expression of ASPM and KIF20A in ESCC and normal tissues adjacent to cancer,and to analyse correlation between their expression and clinicopathological parameters and prognosis.Results Total 16 core genes were screened,including ASPM,KIF20A,etc.GEPIA results showed that ASPM and KIF20A were highly expressed in ESCC tissues with statistically significant difference(P<0.05).KEGG pathway enrichment anal-ysis showed that both of them were correlated with the cell cycle,and TIMER results showed that ASPM was negatively correlated with dendritic cells,and KIF20A was negatively correlated with CD8 + T cells.Immunohistochemical results showed that ASPM and KIF20A in ESCC tissues were significantly higher than those in normal tissues adjacent to the cancer(P<0.001),and ASPM was correlated with clinical stage(P =0.004)and N stage(P =0.024)of ESCC patients,while KIF20A was correlated with the diame-ter of the mass(P =0.015)and N stage(P =0.002)of ESCC patients.Spearman correlation analysis showed that ASPM was posi-tively correlated with KIF20A(r =0.367,P<0.001).Kaplan-Meier survival analysis showed that patients in the group with high expression of ASPM and KIF20A had a lower survival rate than those in the group with low expression(both P<0.05).Multifacto-rial analysis showed that ASPM,KIF20A and N stage were independent factors affecting the prognosis of ESCC patients.Conclu-sion High expression of ASPM and KIF20A is closely related to the progression and prognosis of ESCC,and may be an important prognostic marker and potential therapeutic target for ESCC.
