Danshensu Enhances the Protective Effect of Propofol on Ischemia Reper-fusion Injury in Mice Through Wnt-β-catenin Pathway

本研究基于Wnt-β-catenin通路探讨丹参素增强丙泊酚对小鼠缺血再灌注肾损伤的保护机制.本研究将96只健康雄性小鼠按照随机数字表法分为空白对照组(C组)、假手术对照组(S组)、缺血再灌注组(IR组)、缺血再灌注前进行丙泊酚处理组(IRP组)、缺血再灌注前进行丹参素处理组(IRS组)、缺血再灌注前进行丙泊酚联合丹参素处理组(IRPS组),每组16只.C组正常处理,S组仅游离双侧肾脏,IR组建立小鼠缺血再灌注模型,IRP组于小鼠缺血损伤前30min给予丙泊酚处理,IRS组于小鼠缺血损伤前30min给予丹参素处理,IRPS组于小鼠缺血损伤前30 min给予丙泊酚和丹参素处理,其余处理均与IR组相同.手术2 d后,收集小鼠血清和肾组织,检测血清中肌酐(Cr)和尿氮素(BUN)含量,检测肾组织中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性;HE染色观察肾脏病理学变化;检测肾脏组织Wnt4、β-catenin、Lrp-6的mRNA和蛋白表达水平.结果表明,IR组、IRS组、IRP组、IRPS组的Cr、BUN、MDA水平显著高于C组和S组,且IRPS组的Cr、BUN、MDA水平显著低于IR组、IRP组和IRS组,差异均有显著性(P<0.05);IR组、IRS组、IRP组、IRPS组的SOD活性显著低于C组和S组,且IRPS组的SOD活性显著高于IR组、IRP组和IRS组,差异均有显著性(P<0.05);IR组、IRS组、IRP组、IRPS组的HE染色肾损害程度显著高于C组和S组,且IRPS组的肾损害程度显著低于IR组、IRP组和IRS组,差异均有显著性(P<0.05);IR组、IRS组、IRP组、IRPS组的Wnt4、β-catenin、Lrp-6的mRNA和蛋白水平显著高于C组和S组,IRP组、IRS组、IRPS组的Wnt4、β-catenin、Lrp-6的mRNA和蛋白表达水平显著高于IR组,且IRPS组的Wnt4、β-catenin、Lrp-6的mRNA和蛋白水平显著高于IRP组,差异均有显著性(P<0.05).丹参素可增强丙泊酚对小鼠缺血再灌注肾组织损伤的保护作用,减小肾组织损伤,其机制可能是通过上调Wnt-β-catenin通路达到肾保护作用.

To explore the mechanism of Danshensu enhancing the protective effect of propofol on renal ischemia-reperfusion injury in mice through Wnt-β-catenin pathway.96 healthy male mice were randomly divided into a blank control group(C group),a sham surgery control group(S group),an ischemia-reperfusion group(IR group),a group treated with propofol after ischemia-reperfusion(IRP group),a group treated with Danshensu after ischemia-reperfusion(IRS group),and a group treated with propofol combined with Danshensu after ischemia-reperfusion(IRPS group),with 16 mice in each group.C group was treated normally,while S group only dissociated bilateral kidneys.The IR group established a mouse ischemia-reperfusion model.The IRP group was treated with propofol 30 minutes before ischemic injury,the IRS group was treated with Danshensu 30 minutes before ischemic injury,and the IRPS group was treated with propofol and Danshensu 30 minutes before ischemic injury.All other treatments were the same as the IR group.Two days after surgery,collect mouse serum and kidney tissue,measure the levels of creatinine(Cr)and urinary nitrogen(BUN)in serum,malondialdehyde(MDA)content and superoxide dismut-ase(SOD)activity in kidney tissue;HE staining was used to observe pathological changes in the kidneys;detect the mRNA and protein lelvel of Wnt4,β-catenin,and Lrp-6 in kidney tissue.The results showed that the levels of Cr,BUN,and MDA in the IR group,IRS group,IRP group,and IRPS group were significantly higher than those in the C and S groups,and the levels of Cr,BUN,and MDA in the IRPS group were significantly lower than those in the IR group,IRP group,and IRS group,with significant differences(P<0.05);the SOD activity of the IR group,IRS group,IRP group,and IRPS group was significantly lower than that of the C and S groups,and the SOD activity of the IRPS group was significantly higher than that of the IR group,IRP group,and IRS group(P<0.05);the degree of HE staining renal damage in the IR group,IRS group,IRP group,and IRPS group was significantly higher than that in the C and S groups,and the degree of renal dam-age in the IRPS group was significantly lower than that in the IR group,IRP group,and IRS group(P<0.05);the mRNA and protein levels of Wnt4,β-catenin,and Lrp-6 in IR group,IRS group,IRP group,IRPS group were significantly higher than those in C and S group,the mRNA and protein levels of Wnt4,β-catenin,and Lrp-6 in IRP group,IRS group,and IRPS group were significantly higher than those in the IR group,and the mRNA and protein levels of Wnt4,β-catenin,and Lrp-6 in IRPS group were significantly higher than those in the IRP group(all P<0.05).Danshensu can enhance the protective effect of propofol on renal tissue damage caused by ischemia-reperfusion in mice,reducing renal tissue damage,and its mechanism may be through upregulating Wnt-β-catenin pathway achieves renal protection.
