摘要

Abstract

Objective:To investigate the prognostic value,correlation with occurrence and treatment of circadian rhythm related lncRNA in epithelial ovarian cancer(EOC).Meth-ods:EOC transcriptome data and clinicopathological data were downloaded from the Cancer Ge-nome Atlas(TCGA)and circadian rhythm related genes were downloaded from GeneCards.The risk signature was verified using the Kaplan-Meier method,receiver operating characteristic curve analysis,and concordance index analysis.Based on R language,the relationship between risk model and chemotherapy drug sensitivity was explored,the correlation between risk model and immune function was analyzed,and the nomogram model was constructed.Real-time fluo-rescence quantitative PCR was used to detect the relative expression of lncRNA in EOC tissue and normal ovarian tissue.RT-qPCR was used to investigate the expression of circadian rhythm related lncRNA in EOC tissues.Results:Five of circadian rhythm related lncRNA(AC004540.5、CTD-2013N24.2、FUT8-AS1、CTB-131K11.1、RP11-793H13.8)were selected for establish-ment of the risk model which showed good predictive value in training set or validation set.It was predicted that the drug sensitivity of paclitaxel,docetaxel and cyclophosphamide was closely related with high risk,and the drug sensitivity of irinotecan and topotecan was related with low risk.Enrichment analysis revealed that the risk model was closely related to immune-related ac-tivity.RT-qPCR results showed that the expression differences of AC004540.5,CTD-2013N24.2,FUT8-AS1,CTB-131K11.1 and RP11-793H13.8 between normal and cancer tissue were consistent with the results of big data analysis.Conclusions:A risk model based on five circadi-an rhythm related lncRNA with prognostic value may be a promising prognostic tool in EOC and guide individualized patient treatment.
