Effect of human umbilical cord mesenchymal stem cell transplantation via caudal vein on apoptosis-related proteins in ovaries of perimenopausal rats

Abstract
Objective:To explore the improvement of ovarian function in perimenopaus-al rats after transplantation of human umbilical cord mesenchymal stem cells(hUCMSCs).Methods:Thirty female rats aged 13~15 months were selected to construct a perimenopausal rat model.Perimenopausal mice were divided into hUCMSCs transplantation group and blank group,with 15 mice in each group.Another 15 healthy female rats were selected as the young control group.Before hUCMSCs transplantation,venous blood was collected from rats in each group to detect serum E2,AMH,and FSH levels.2 weeks,3 weeks,and 4 weeks after hUCMSCs transplantation,5 rats from each group were randomly selected to detect serum E2,AMH,and FSH,and ovarian tissue was collected to detect Bcl-2.Bax and Caspase-3 protein expression levels.Results:After hUCMSCs transplantation,the E2 and AMH of the rats in the hUCMSCs group were higher than those of the blank group,and the FSH was lower than that of the blank group,and the differences were statistically significant(P<0.05).The ovarian primordial folli-cles,primary follicles,and secondary follicles of rats in the hUCMSCs group,the number of an-tral follicles in the hUCMSCs group was higher than that in the blank group,and the difference was statistically significant(P<0.05).4 weeks after transplantation,the expressions of Bcl-2 and Bcl-2/Bax in the hUCMSCs group were higher than those in the blank group,and the ex-pressions of Bax and Caspase-3 were lower than those in the blank group,the difference was sta-tistically significant(P<0.05).Conclusion:After the transplantation of hUCMSCs,levels of E2 and AMH in rats increased while FSH decreased.Additionally,the increase in ovarian Bcl-2 and Bcl-2/Bax ratio as well as the decrease in Bax and Caspase-3 further supports the potential role of hUCMSCs in regulating the process of ovarian cell apoptosis.关键词
