Effects of Lactobacillus rhamnosus on gut flora and offspring of the rats with advanced maternal age

Abstract
Objective:To explore the effect of Lactobacillus rhamnosus(LGG)on gut flora and the offspring.Methods:10 SPF-grade 3-month-old SD female rats(non-advanced ma-ternal age,NMA)and 8 12-month-old SD female rats(advanced maternal age,AMA)were se-lected and caged with 3-month-old SD male rats in a ratio of 2 ∶ 1,and were divided into four groups after cage mating:non-advanced maternal age control group(NMA.C group),non-ad-vanced maternal age intervention group(NMA.E group),advanced maternal age control group(AMA.C group)and advanced maternal age intervention group(AMA.E group).The control groups were given saline 0.25mL/day by intragastric administration,and the intervention groups were given LGG preparation 1×109 CFU/(0.25mL·day)by intragastric administration for 14 days,and then collected the feces,serum and placenta of pregnant rats.Results:Compared with the AMA.C group,the activity of superoxide dismutase(SOD)in placenta of the AMA.E group increased,and the serum pro-inflammatory cytokines IL-6 and TNF-α decreased,while anti-in-flammatory cytokine IL-10 increased.The abundance of Lactobacillales and Lactobacillaceae in the AMA.E group was significantly higher than that in the AMA.C group,while the abundance of Clostridia in the AMA.C group was significantly higher.The number and length of offspring increased significantly in the AMA.E group.Conclusion:The results showed that supplementa-tion of LGG during pregnancy could improve the antioxidant level of pregnant rats and reduce inflammatory reaction,increase the species abundance and uniformity of intestinal flora and the abundance of beneficial bacteria,reduce the abundance of pathogenic bacteria,and improve the intestinal flora disorder of the rats with AMA to varying degrees.关键词
