外源性白细胞介素10抑制嗜酸性粒细胞型慢性鼻窦炎伴鼻息肉中白细胞介素33表达OACSTPCD
Exogenous IL-10 inhibits the expression of IL-33 in eosinophilic chronic sinusitis with nasal polyps
目的 研究慢性鼻窦炎伴鼻息肉(CRSwNP)患者鼻息肉中白细胞介素10(IL-10)、IL-33的表达水平,及外源性IL-10刺激对嗜酸性粒细胞型CRSwNP(eosinophilic CRSwNP,ECRSwNP)中IL-33表达水平的影响.方法 手术获取ECRSwNP 12例、非嗜酸性粒细胞型CRSwNP(non-eosinophilic CRSwNP,nECRSwNP)8例及12例正常鼻黏膜组织,采用酶联免疫吸附法(ELISA)测定组织中IL-10及IL-33表达水平.另收集10例ECRSwNP样本,以含或不含100 ng/ml IL-10的培养基分别培养同一患者样本24 h后,ELISA及免疫组化检测IL-33表达量.结果 IL-10在ECRSwNP组、nECRSwNP组、对照组中表达无显著差异(P>0.05).IL-33在ECRSwNP组中的表达水平高于对照组,差异有统计学意义(P=0.0043).在ECRSwNP组中,IL-10与IL-33表达水平呈显著负相关(r=-0.7470,P=0.0052);在nECRSwNP组及对照组中两者无明显相关性(P>0.05).相比于空白对照组,IL-10刺激组中IL-33的表达显著下降(P=0.0197)、免疫组化观察到IL-33阳性细胞显著减少(P=0.0183).结论 在ECRSwNP中IL-33的表达量显著增高,IL-10与IL-33负相关.外源性IL-10可以抑制ECRSwNP中IL-33的表达.
OBJECTIVE To investigate the expression levels of interleukin-10(IL-10)and IL-33 in patients with chronic rhinosinusitis with nasal polyps(CRSwNP)and the effect of exogenous IL-10 stimulation on the expression level of IL-33 in eosinophilic CRSwNP(ECRSwNP).METHODS The expression levels of IL-10 and IL-33 in nasal polyp stissues of 12 patients in ECRSwNP group,8 patients in non-eosinophilic CRSwNP(nECRSwNP)group,and normal nasal mucosa of 12 healthy controls were determined by ELISA.Another 10 ECRSwNP samples were collected and cultured in medium with or without 100 ng/ml IL-10 for 24 hours,and then the expression of IL-33 was detected by ELISA and immunohistochemistry.RESULTS There was no significant difference in expression of IL-10 among ECRSwNP group,nECRSwNP group and healthy control group(P>0.05).The expression level of IL-33 in ECRSwNP group was higher than that in healthy control group,and the difference is statistically significant(P=0.0043).The expression level of IL-10 and IL-33 was a significant negative correlation in ECRSwNP group(r=-0.7470,P=0.0052).There was no significant correlation in nECRSwNP group and control group(P>0.05).Compared with the control group,the expression level of IL-33 in the IL-10-stimulated ECRSwNP group was significantly decreased(P=0.0197),and a significant decrease of IL-33 positive cells was observed in immunohistochemistry(P=0.0183).CONCLUSION IL-33 expression is significantly increased in ECRSwNP,and IL-10 is negatively correlated with IL-33.Exogenous IL-10 can inhibit IL-33 expression in ECRSwNP.
