Exogenous IL-10 inhibits the expression of IL-33 in eosinophilic chronic sinusitis with nasal polyps

Abstract
OBJECTIVE To investigate the expression levels of interleukin-10(IL-10)and IL-33 in patients with chronic rhinosinusitis with nasal polyps(CRSwNP)and the effect of exogenous IL-10 stimulation on the expression level of IL-33 in eosinophilic CRSwNP(ECRSwNP).METHODS The expression levels of IL-10 and IL-33 in nasal polyp stissues of 12 patients in ECRSwNP group,8 patients in non-eosinophilic CRSwNP(nECRSwNP)group,and normal nasal mucosa of 12 healthy controls were determined by ELISA.Another 10 ECRSwNP samples were collected and cultured in medium with or without 100 ng/ml IL-10 for 24 hours,and then the expression of IL-33 was detected by ELISA and immunohistochemistry.RESULTS There was no significant difference in expression of IL-10 among ECRSwNP group,nECRSwNP group and healthy control group(P>0.05).The expression level of IL-33 in ECRSwNP group was higher than that in healthy control group,and the difference is statistically significant(P=0.0043).The expression level of IL-10 and IL-33 was a significant negative correlation in ECRSwNP group(r=-0.7470,P=0.0052).There was no significant correlation in nECRSwNP group and control group(P>0.05).Compared with the control group,the expression level of IL-33 in the IL-10-stimulated ECRSwNP group was significantly decreased(P=0.0197),and a significant decrease of IL-33 positive cells was observed in immunohistochemistry(P=0.0183).CONCLUSION IL-33 expression is significantly increased in ECRSwNP,and IL-10 is negatively correlated with IL-33.Exogenous IL-10 can inhibit IL-33 expression in ECRSwNP.关键词
